## Overview

SLU-PP-332 is an investigational small-molecule agonist of the estrogen-related receptors (ERRalpha, ERRbeta, ERRgamma), a family of orphan nuclear receptors that regulate metabolic and mitochondrial gene programs. By activating these receptors, SLU-PP-332 is proposed to mimic the metabolic effects of exercise, including enhanced fatty acid oxidation, improved mitochondrial function, and increased endurance. It is studied as an exercise mimetic for metabolic disease and performance. SLU-PP-332 has an estimated half-life of roughly five hours and about 50% bioavailability. The evidence base is almost entirely preclinical, with the supporting data coming from cell and rodent studies that showed improved exercise capacity and metabolic markers. Human pharmacokinetic and efficacy data does not exist, and SLU-PP-332 is not an approved medication.
