## Overview

Retatrutide is an investigational once-weekly peptide that acts as a triple agonist of the GLP-1, GIP, and glucagon receptors, in development for obesity and type 2 diabetes and notable for the magnitude of weight loss reported in Phase II data. By co-activating GLP-1 and GIP receptors it drives insulin release, slows gastric emptying, and reduces appetite, while the glucagon receptor activity adds direct hepatic fat oxidation and thermogenesis that dual GLP-1/GIP agents do not provide. The compound has a roughly 147-hour half-life, peaks within roughly 1.7 days of subcutaneous injection, and shows about 80% bioavailability, which is what allows once-weekly dosing with sustained exposure over months. Phase II data show weight loss exceeding current GLP-1 and GLP-1/GIP comparators, while the dominant adverse effects are gastrointestinal, including nausea, vomiting, diarrhea, and reduced appetite, alongside the known class considerations around lean mass preservation.
