## Overview

Pramlintide (Symlin) is a synthetic analog of amylin, the peptide hormone co-secreted with insulin by pancreatic beta cells. It works at mealtimes to slow gastric emptying, suppress postprandial glucagon, and promote satiety, complementing insulin glucose-lowering effects. It is approved as an adjunct to mealtime insulin in type 1 and type 2 diabetes and is also studied for weight management. Pramlintide has a short half-life of roughly 50 minutes and about 40% bioavailability, so it is injected before meals rather than as a long-acting depot. Pramlintide is an FDA-approved medication with solid clinical evidence in diabetes. The main practical constraints are the short half-life requiring multiple daily injections and the risk of hypoglycemia when combined with insulin.
