## Overview

MOTS-c is a mitochondrial-derived peptide encoded by a short open reading frame in mitochondrial DNA, specifically within the 16S rRNA gene. It acts as a signaling molecule that regulates metabolic homeostasis, including AMPK activation, improved insulin sensitivity, and the upregulation of antioxidant defenses. It is studied as an exercise mimetic and metabolic regulator. MOTS-c has an estimated half-life of about 12 hours and around 50% bioavailability. Most of the supporting evidence comes from cell and animal work, with human pharmacokinetic data very limited. A few small human studies have begun, but MOTS-c is not an approved medication, and the dosing comes from preclinical extrapolation.
