## Overview

Mechano growth factor (MGF) is a splice variant of IGF-1, specifically the IGF-1Ec isoform, that skeletal muscle produces in response to mechanical load and damage. Unlike the systemic IGF-1Ea variant, MGF acts locally, activating satellite cells and triggering muscle repair signaling within hours of injury. Its distinct C-terminal E-domain drives these regenerative effects and separates it from other IGF-1 splice forms. The peptide clears fast, with an estimated half-life of roughly 30 minutes and around 50% subcutaneous bioavailability. Research contexts typically reference 200 to 800 mcg doses, but these numbers come from preclinical work rather than controlled human trials. Human pharmacokinetic data is essentially absent, and MGF is not an approved medication. Because native MGF degrades in minutes, researchers often turn to PEG-MGF for a longer-acting alternative.
